Genetic Diversity and Relationship of Durian (Durio spp.) Germplasm Based on the Internal Transcribed Spacer (ITS) Region: In Silico Analysis

Durian (Durio spp.) is a germplasm with relatively high species diversity, an estimated 27 worldwide. However, the existence of several has been threatened. This study aimed to reconstruct DNA barcode durian and its relatives analyze genetic diversity relationship based on internal transcribed spacer (ITS) region. Sixteen sequences durians ITS were collected from GenBank (NCBI) analyzed in silico using BLAST, MultAlin, MEGA-X software, then reconstructed phylogenetically by UPGMA Maximum Likelihood methods. The results show that region Durio spp. base length about 702 bp, where mutations occur, substitution (transversion transition) indel (insertion deletion). At nucleotide level, this shows 0.065. cluster analyses (UPGMA Likelihood) can separate into four clusters five main clades, respectively. In study, D. zibethinus, most popular genus, closely related lowianus far griffithii. information beneficial as reference data support conservation breeding programs, locally globally, especially Indonesia.
